Ink Gestures for Word
Loren (one of at least three Heiny's) over at Incremental Blogger is working on a plug-in for Microsoft Word that will allow you to use common proofreader's marks to make actual changes to Word Documents. There are some animated screenshots at his site that show the functionality in action.
For someone very familiar proofreading marks already, this could certainly qualify as a killer app. Personally, I'm not sure how much I'd use it, since having a convertible means that I'd probably do most of my editing with the keyboard. Still, this is the sort of thing that could convert someone who is used to marking up papers in this manner.

However, I'm still of a mind that the first true killer app for the Tablet will be the only that breaks the "pen and ink" motif and uses the stylus for something truly original (or at least, less literal).
